So between them, Plasmoid is probably overall best for advantage and resistances, Thri Kreen is best if you are okay with going for either a light weapon and shield, or dual wielding without a shield, since you can still grapple with one free arm that way. Advantage on Str saving throws is actually nice, although if starting Fighter you'll already probably have a high score and proficiency. Giff also get the advantage on strength checks. Their psudeopod limb also doesn't let them grapple. They've also got some good resistances and utility. You won't have Giants Might all the time so it's still useful. Thats good, but Rune Knight already has a way to get that. Plasmoids have advantage to initiate or escape grapples. But being stuck with light weapons is a big deal in a fighter. So they'd be great for being able to grapple, use a shield, use a weapon and have a free hand. Thri Kreen can't grapple or use a shield with their arms, but they can use a light weapon. If you want a support/grappling tank that can also deal damage, go for Rune Knight. If you want a ranged/throwing damage dealer, go for Giant Barb. Protecting your allies, locking down enemies, and forcing them to lose saves or help your allies make them. Rune Knight gets multiple spell like abilities that are incredibly powerful supports. It's got fantastic forced movement to keep distance and can work well with allies AOE effects, like Web and Spike Growth. Barbarian will do better weapon damage than RK at least until level 11, since it's stacking multiple sources of extra damage with Reckless Attack. Giant is a Strength range attacker, than can also attack in melee. So actually they're very different in play. Barbarian gets it more often and earlier. Rune Knight gets damage resistance once a short rest at level 7. Rune Knight gets lots of skill ability boosts, including Darkvision and no longer being suprised. As a bonus action, Giant can throw people 30 feet. Giant gets bonus regular damage to thrown weapons and elemental damage for each hit, with regular weapons becoming thrown as well. RK gets bonus damage only once per turn. It's one of the best abilities in the game. Specifically things like Cloud Rune, which simply redirects an enemy hit into another combatant, without a save, and Storm Rune, with lets you impose advantage/disadvantage with a reaction for a whole minute, including on enemies when your spellcaster ally uses a save or suck.
